I have used a Rotary Microtome to section resin-embedded charcoal samples. I was able to get sections that were larger than those obtained with a Sliding Microtome. I kept the Knife and Block Face wet with 70% ethanol during sectioning. I cut sections slowly and cushioned the sections with a wet brush while they were being cut. I learned a long time ago that you need to improvise a bit to get good sections. Sections are placed in water drops on a microtome slide. These are placed on a slide warmer set to 60 degrees C. This expands the resin and removes wrinkles. It also helps them adhere to the slide. Sections can be photographed immediately by adding water and a coverslip. Permanent mounts are made by adding a Mounting Medium like Polymount (Polysciences) or Permount. After applying a coverslip, lead weights (bullet tips) are used to press the coverslip down. These can be revoved after a few days. Excess resin can be removed with a razor blade. |
