Stems are the easiest specimens to sample. It is important to realize that you do NOT need complete cross-sections of uniform thickness. Stems have radial symmetry. Consequently, one sector of the stem may be all that you need to document the anatomy or to identify specific structures and cell types. The best results occur when fresh teflon-coated blades are used. An old fashioned straight razor would make a good field microtome blade that could be used over and over again. These are exceedingly sharp and can be easily maintained. Teflon-coated razor blades are common place. The best blades are those designed for old-fashioned double-edged shavers. These are very very sharp & must be handled with great care. You might apply tape to one of the edges to avoid cutting yourself.Teflon sprays can be purchased so that blades can be coated immediately prior to use. |
