The Nucleus is the largest substantial organelle. It is translucent and is hard to observe in living material unless you use specialized microscopes. This cell was stained with Toluidine Blue. The Nucleus & Nucleolus are evident. Chloroplasts are barely visible.

This shows a Nucleus & Nucleolus viewed with Phase Contrast microscopy. The streaks are due to organelles in the cytoplasm.
